ในเกม The Spirit of the Samurai ผู้เล่นจะได้รับบทเป็น "Takeshi" ซามูไรหนุ่มผู้มีหน้าที่ปกป้องหมู่บ้านจากการรุกรานของเหล่าปีศาจ "Oni" และเหล่ากองทัพปีศาจที่ต้องการยึดครองโลก โดยมี "Chisai" แมวผู้กล้า และ "Kodama" วิญญาณตัวจิ๋ว คอยเป็นเพื่อนร่วมทาง โดยผู้เล่นจะได้ออกผจญภัยในโลกที่ได้รับแรงบันดาลใจมาจากตำนานและนิทานพื้นบ้านของญี่ปุ่น อาทิ หมู่บ้านร้าง ถ้ำบนภูเขา สุสานร้าง และสถานที่อื่น ๆ อีกมากมาย พร้อมเผชิญหน้ากับเหล่ายูกิ (Yokai) สัตว์ประหลาด และปีศาจต่าง ๆ ที่ถูกสร้างสรรค์ขึ้นมาอย่างสวยงามในสไตล์สต็อปโมชั่นแบบ Ray Harryhausen

นอกจากนี้ผู้เล่นจะได้สลับสับเปลี่ยนระหว่างการเล่นด้วยตัวละครทั้งสาม ได้แก่ Takeshi ซามูไรผู้เชี่ยวชาญการต่อสู้ด้วยดาบ, Kodama วิญญาณตัวจิ๋วที่มีความสามารถในการปีนป่าย และ Chisai แมวผู้กล้า ที่แต่ละตัวจะมีวิธีการโต้ตอบกับสิ่งแวดล้อมที่แตกต่างกัน อีกทั้งผู้เล่นจะได้ใช้ดาบคาตานะ หอก และธนู ซึ่งเป็นอาวุธโบราณของญี่ปุ่น เพื่อต่อสู้กับเหล่าปีศาจ นอกจากนี้ยังมีท่าไม้ตายและคอมโบสุดอลังการที่รอให้ผู้เล่นได้ปลดล็อคและพัฒนา

ตลอดการเดินทางผจญภัยผู้เล่นได้รับประสบการณ์ที่ช่วยให้พัฒนาทักษะและความสามารถต่าง ๆ ได้อย่างมากมาย ไม่ว่าจะเป็นการเพิ่มพลัง เพิ่มค่าสถานะ หรือเรียนรู้ท่าไม้ตายใหม่ ๆ เพื่อเอาชนะศัตรูที่แข็งแกร่งยิ่งขึ้น
